Effect of Diptheria Toxin on Growth of Sarcoma-180 in Mice

Abstract
In order to prove the anti-tumor activity of the diphtheria toxin in mice bearing sarcoma-180 ascites tumor, the survival times of various experimental groups treated with various doses of toxin, treated at stages of tumors, treated by intermittent administrations, and treated with fractionated or single large doses, were investigated. The results were as follows:
1. In the control mice with sarcoma-180 ascites tumor treated by saline once daily for 2 consecutive days beginning one day after the transplantation of tumor, the mean survival time was 14.6 days. The survival times for the groups treated by diphtheria toxin of 1 MLD/mouse/day, 3 MLD/mouse/¡©day,5 MLD/mouse/day and 7 MLD/mouse/day for 2 consecutive days beginning one day after trans¡©plantation were 16.8 days(15. 1% increased than saline control), 24. 1 days (65. 1%, increased), 25.6 days(68. 5o increased), and 23. 1 days (58.2% increased), respectively.
2. The survival time of control group treated by toxin of 3 MLD/mouse/day for 2 consecutive days beginning one day after transplantation was 23.9 day. In other groups treated by toxin of 3 MLD/ mouse/day for 2 consecutive days beginning three and five days after the transplantation of tumor, the survival times were 21. 7 days (9. 2% decreased than control), and 15. 3 day (56.2%, decreased), respectively.
3. The survival time of control group treated by toxin of 3 MLD/mouse/day for 2 consecutive days beginning one day after transplantation was 23.3 days. In various groups treated by toxin of 3 MLD /mouse/day for 2 intermitent days on the 1st and 3rd days, 1st and 5th days, and 1st and 7th days after tumor transplantation, the survival times were 20. 2 days(13.3% decreased than control), 17. 9 days (23.2% decreased), and 16. 7 days(28. 3% decreased), respectively.
4. The survival time of control group treated by toxin of 3 MLD/mouse/day for 2 consecutive days beginning one day after transplantation was 23. 3 days. In various groups treated by toxin of 2 MLD/mouse/day for three days on the 1st, 2nd and 3rd days, the lst, 3rd and 5th days, and the 1st, 5th and 7th days after tumor transplantation, the survival times were 21.6 days(9. 6% decreased than control), 18. 2 days(32. 2% decreased), and 17. 8 days(25. 5% decreased), respectively.
5. The survival time of control group treated by toxin of 3 MT D/mouse/day for 2 consecutive days beginning one day after transplantation was 23.3 days. In various groups treated by toxin of 6 MLD /mouse/day for one day on the Is¢¥, 3rd, 5th and 7th days after tumor trasplantation, the survival times were 16. 9 days(decreased 29. 3% of control), 16. 1 days(decreased 32. 6%), 14. 4 days(39. 3% decreased) and 13. 9 days(decreased 41. 2%), respectively.
It was concluded that diphtheria toxin prolonged the survival times of mice bearing sarcoma-180 ascites tumor in this experiment.
